Hyperpolarization-activated cyclic nucleotide-gated (HCN) channels contribute to a wide range of physiological functions and regulate cardiac and neuronal pacemaker activity. Here, we review recent advances in three areas: the polarity of the S4 movement with hyperpolarization, the location of the activation gate, and the structure of the C-terminal C linker and CNBD.
